Substantiation Nirogacestat zal waarschijnlijk in de plaats komen van sorafenib. Het liet een significante PFS verbetering vergeleken met placebo zien (hazard ratio [HR], 0.29 [95% CI, 0.15, 0.55; P<0.001]). De ORR was ook significant verbeterd met nirogacestat versus placebo (41% versus 8%; P<0.001), met een mediane tijd tot respons van 5,6 versus 11,1 maanden. De complete response rate was 7% met nirogacestat versus 0% met placebo. DFS voldoet aan PASKWIL.

Additional remarks DT's beslaan <3% van de wekedelentumoren (1). In 2019 werden er in Nederland 768 wekedelensarcomen gediagnosticeerd (2). Dit zou betekenen dat er minder dan 23 patiënten per jaar met desmoïd tumoren zouden worden gediagnosticeerd. Daarnaast wordt de jaarlijkse incidentie geschat tussen 1 op 250.000 en 1 op 500.000 (1). Voor Nederland zou dit 34 tot 68 patiënten betekenen. De daadwerkelijke incidentie zal daarom waarschijnlijk enkele tientallen bedragen.
