Additional remarks Risdiplam heeft recentelijk PRIME designation ontvangen van EMA. In de loop van dit jaar start er een extended access studie (NCT04256265). De fabrikant verwacht registratie in april 2021. Sinds 23 oktober 2020 is er een compassionate use programma toegekend door het CBG.

Substantiation Risdiplam heeft goede resultaten op de motorfunctie. Daarnaast is een groot voordeel dat dit middel oraal kan worden toegediend. Net als nusinersen heeft het geneesmiddel het grootste effect bij jonge kinderen. In de extended access studie zijn positieve resultaten gevonden voor SMA type 1 en 2. Voor SMA type 3 zullen de definitieve resultaten later gepubliceerd worden.

Additional remarks Nusinersen wordt voor een brede indicatie zoals hierboven beschreven ingeschat. Het patiëntvolume is op dit moment onbekend, omdat dit afhankelijk zal zijn van de precieze indicatie en studieresultaten. Van 327 SMA patiënten is het type bekend:190 hiervan hebben SMA type 1 of 2, de overige 137 hebben SMA type 3. Door de extended access studie zal risdiplam voor een grotere populatie patiënten beschikbaar komen. Op dit moment zullen in ieder geval patiënten met SMA type 1 en 2 in aanmerking komen en bij positieve resultaten op stabilisatie van motorische functies mogelijk ook patiënten met type 3.
