Substantiation In de CAPItello-281 fase 3 studie werd onderzocht of toevoeging van capivasertib aan abirateron effectief is bij patiënten met PTEN-deficiënte gemetastaseerde hormoongevoelige prostaatkanker. Van de 6003 patiënten met een geldig tumortestresultaat had 25,3% PTEN-deficiëntie. In deze groep leidde capivasertib plus abirateron tot een statistisch significante verbetering van de radiografische progressievrije overleving (rPFS): mediaan 33,2 maanden versus 25,7 maanden bij placebo plus abirateron (HR 0,81; P = 0,034). Post-hoc analyses toonden aan dat het behandelingsvoordeel toenam bij hogere PTEN-verliesdrempels. Voor de totale populatie was er geen significant verschil in algehele overleving. Veelvoorkomende bijwerkingen van capivasertib waren diarree (51,9%), hyperglykemie (38,0%) en huiduitslag (35,4%). Het aantal sterfgevallen gerelateerd aan bijwerkingen was iets hoger in de capivasertib-groep (7,2% versus 5,2%). Omdat er geen significante algehele overleving winst is, is het onwaarschijnlijk dat dit aan de PASKWIL-criteria gaat voldoen.
