Substantiation In 2022 werd crizotinib geregistreerd voor de leeftijdsgroep 6 jaar en ouder. Dit was de eerste geregistreerde ALK-remmer voor kinderen in Europa. Xalkori werd onderzocht bij 22 patiënten met ALCL & 14 patiënten met IMT. De ORR was 86% voor ALCL & 86% voor IMT. Deze indicatie uitbreiding is bekostigd als add-on per 1 december 2024. De aanbevolen dosering van crizotinib voor pediatrische patiënten met ALCL of IMT is 280mg/m2 oraal tweemaal daags tot progressie van de ziekte of onaanvaardbare toxiciteit, in de SmPC wordt een doseringsschema gegeven. Additional remarks Jaarlijks zijn er 35 tot 50 kinderen met een non-Hodgkin lymfoom, daarvan heeft 10 tot 20% ALCL (3 tot 10 kinderen). Van deze 3 tot 10 kinderen is 80% ALK+ (3-8 kinderen), waarvan 25 tot 40% relapsed/refractory is (1 tot 3 kinderen). Maximaal komen er per jaar 1 tot 3 kinderen met ALCL in aanmerking voor behandeling met crizotinib. IMT Jaarlijks zijn er 40 kinderen met een wekedelen tumor, daarvan heeft 5% IMT (2 kinderen). Van deze 2 kinderen is 66,7% ALK+ (1-2 kinderen), waarvan 30 tot 50% unresectable, recurrent of refractory is (0 tot 1 kinderen). Maximaal komen er per jaar 0 tot 1 kinderen met IMT in aanmerking voor behandeling met crizotinib. In totaal gaat het jaarlijks voor beide indicaties om 1 tot 4 kinderen die behandeld kunnen worden met crizotinib. Alle patiënten worden behandeld in het Prinses Máxima centrum.
