Substantiation In de gepubliceerde resultaten van de fase 3 studie (AMPLITUDE) werd onderzocht of niraparib gecombineerd met abirateron en prednison (AAP) effectief is bij patiënten met metastatische castratiegevoelige prostaatkanker (mCSPC) met HRR-genafwijkingen, zoals BRCA1/2. De combinatie leidde tot een significante verbetering van de radiografische progressievrije overleving (rPFS), vooral bij patiënten met BRCA-mutaties (HR = 0,52; P < 0,0001) en ook in de totale populatie (HR = 0,63; P = 0,0001). De gegevens over algehele overleving (OS) zijn echter nog onvolledig. Hiervan zal circa 30% een afwijking hebben in HRR-genen (n=626). In Nederland worden mannen met prostaatkanker echter pas laat in het behandeltraject op mutaties getest na falen van standaardbehandeling(en). Het daadwerkelijke aantal patiënten met mHSPC waarvan de HRR-mutatiestatus bekend is en die in aanmerking zullen komen voor niraparib/abirateron, zal dus veel kleiner zijn.
