Door de werking van a-MSH na te bootsen, stimuleert afamelanotide melanocyten om de productie en afgifte van melanine te verhogen. Verhoogd melanine in melanocyten kan bescherming bieden tegen door ultraviolette straling (UVR) geïnitieerde cellulaire DNA-schade, oxidatie van membraaneiwitten en veranderingen in intracellulaire signaalprocessen in epidermale cellen. Endogeen wordt a-MSH afgegeven door huidcellen in reactie op blootstelling aan UVR, waardoor melanocyten gestimuleerd worden melanine te produceren en af te geven.
