Substantiation Aficamten lijkt vergelijkbaar te zijn met mavacamten. Er zijn wel een aantal verschillen in product eigenschappen, zoals de halfwaardetijd, het is farmakinetisch iets voorspelbaarder en er lijken minder veiligheidsproblemen. In de fase 3 was er een duidelijk effect vergeleken met bèta blokkers en calcium antagonisten bij een selecte populatie. Er zal in de praktijk gekeken moeten worden of mavacamten of aficamten de voorkeur krijgt. Echter, worden 6 van de 7 HCM patiënten niet gediagnosticeerd of mis-gediagnosticeerd volgens studies. Ongeveer 30% van de gediagnosticeerde HCM patiënten hebben obstructieve HCM n=1.209 (4). Van de populatie patiënten met obstructieve HCM, heeft zo'n 86% NYHA functionele klasse II en III in aanmerking n=1.039 (5).
